I have read the comments here and I agree with most of the users on here. Althought I am straight, many people have doubted my sexuality, (assuming I was a lesbian because I have predominently female friends.)

My thing is if it is someone I know, I tell them to pay attention to my actions and conversations then get back to me.

As for those that I don't know? I make a quick witted remark to get them away from me. When it comes to sexuality, I don't judge and it is not a sensitive topic for me. But, when it is someone I don't know, like or has malicious intents I amd known for being rather sharp tongued.

Normally, if you are assertive and don't back down when confronted by someone you aren't comfortable with this question they back down and get the hint to never ask you that question again. That works for me and my peers.
